Here’s a nice article describing what your page should look like when you’re targeting the Web 2.0. Besides making heavy use of AJAX, you should take in attention rounded shapes, shades or bigger fonts.
One aspect that was not referred but I think it should be highlighted: following web standards. Nowadays is absolutely critical that you follow the W3C web standards!
